While biotechnological advances, genomics and high throughput screenings or combinatorial and asymmetric syntheses are opening new opportunities in drug discovery, the industry is facing serious innovation deficit. The total number of new molecules registered per year has dropped in contrast to expected increase. Post marketing failures of blockbuster drugs have become major concerns of industries. On the other side, globally there is a major shift to sue of traditional medicine involving complementary and alternative therapies. Ethnopharmacology and traditional medicines have contributed in past significantly in the process of natural product drug discovery. There are two clear tracks where ethnopharmacology has potential to contribute in future drug research. First, as a discovery engine to provide new targets, leads, and second, use of quality assured and standardized traditional medicines. In this scenario, it is important to understand the mechanisms of drug discovery and pharmaceutical development with a focus on herbal drugs and neutraceutical. This book provides historical perspective, future prospects and significance of ethnopharmacology in drug research. It also provides important steps in botanical drug discovery and development including bioprospecting, quality control, standardization, pharmaceutics, stability, pharmacokinetics, and bioavailability with examples from ethnopharmacology and herbal medicine. One of the important feature of this book is to give an excellent insight to Good Laboratory and Good Clinical Practices along with very useful summary steps involved in filing IND or NDA of botanical products. The book also gives Regulators' perspective of validating claims and how ethnopharmacological or traditional medicines need different approach.

Herbs and herbal products are of paramount importance for human health. To beable to guarantee safety and quality, standards and testing methods are needed.Pharmacopoeias contain quality control protocols setting the standards which arethen required by governments. The quality traits are many, including the intrinsicvariables of medicinal plant, e.g. the levels of the active compounds, and the absenceof possibly natural occurring toxic compounds. On the other hand, many qualitytraits are related to agricultural conditions and practices, or to the harvesting andpost-harvest processing. With so many variables, quality control of the end productbecomes extremely complex, time consuming and costly. To ensure the quality ofmedicinal plants for human consumption quality management -the use of “goodpractices” at each step, from seed to final product- becomes a crucial aspect. In general, quality control includes the inspection of the product’s identity, purity, and content, based on its physical, chemical or biological properties. To ensure thequality of herbal medications, criteria such as botanical quality, type of preparation, physical constants, adulteration, contaminants, chemical constituents, pesticidesresidues et al. should be examined. Meanwhile, authentication of herbs is neededto avoid possible adulteration or contaminating plants, even toxic herbs such asAristolochia species. Many of the methods are long standing, such as microscopy incombination with color reactions, but some 50 years ago chromatography developedas a major tool for both qualitative and quantitative analysis of herbal preparations.Nowadays, research is working on the improvement of these methods and on thedevelopment of novel tools. For instance, next generation sequencing and mass spectrometry imaging, areemerging as new technologies for the quality control of herbal medicines. Withthese technologies, quick testing of herbal products and of mixed herbal powderpreparations, including the testing for specific plant parts (botanical drugs), can beachieved. Also, novel chemical tools such as metabolomics and Near Infrared Red(NIR) spectroscopy are being developed as powerful tools to identify and to linkthese with activity by using chemometric tools such as multivariate analysis. Finally, progress of informatic tools such as machine learning helps to deal with the big datagenerated by sequencing or mass spectrometry. However, these new technologies, like all other new born technologies, should be tested and perfected for a broaderrange of products.

"This book presents a structural approach to the evaluation of herbal medicinal products for quality, safety and efficacy. There has been an enormous growth in the market for herbal medicinal products in the last twenty five years. However the rediscovery of natural substances with therapeutic potential has raised questions of quality, safety and efficacy on the part of the consumer and also from health professionals. This book brings together current thinking and practice in these areas highlighting current research. In the light of increasing legislation to enforce better standards for these products and the demand by legislators and the public for assurance of safe and effective use, this book seeks to provide a state-of-the-art review, which informs and guides those who seek to promote their use. This book also gives an overview of the place of ethno pharmacology in the development of herbal medicinal products and discusses good agricultural and collection practices, marker analysis and stability testing which contribute to assessment of good quality of these materials." -- Publisher description.

Medicinal plant materials are supplied through collection from wild populations and cultivation. Under the overall context of quality assurance and control of herbal medicines WHO developed the Guidelines on good agricultural and collection practices (GACP) for medicinal plants providing general technical guidance on obtaining medicinal plant materials of good quality for the sustainable production of herbal products classified as medicines. These guidelines are also related to WHO's work on the protection of medicinal plants aiming promotion of sustainable use and cultivation of medicinal plants. The main objectives of these guidelines are to: (1) contribute to the quality assurance of medicinal plant materials used as the source for herbal medicines to improve the quality safety and efficacy of finished herbal products; (2) guide the formulation of national and/or regional GACP guidelines and GACP monographs for medicinal plants and related standard operating procedures; and (3) encourage and support the sustainable cultivation and collection of medicinal plants of good quality in ways that respect and support the conservation of medicinal plants and the environment in general. These guidelines concern the cultivation and collection of medicinal plants and include certain post-harvest operations. Good agricultural and collection practices for medicinal plants are the first step in quality assurance on which the safety and efficacy of herbal medicinal products directly depend. These practices also play an important role in protection natural resources of medicinal plants for sustainable use.

This book provides a step-by-step guide to simple methods for verifying the identity of commonly used pharmaceutical substances and dosage forms. The basic tests described can also be used to detect mislabeled, substandard, or counterfeit products when the labeling or physical attributes give rise to doubt. Intended for use in developing countries, where resources and specialized skills may be scarce, all tests rely on a limited range of easily available reagents and equipment and need not be performed in a fully equipped laboratory or by persons with specialized training in pharmacy or chemistry. The book describes tests for 23 pharmaceutical substances and 58 pharmaceutical dosage forms, most of which are included in the WHO Model List of Essential Drugs. Basic tests for confirming the identity of four commonly used medicinal plant materials are also included. As stressed in the text, these tests, which merely confirm identity, are intended for use as primary screening tools and may need to be followed, in cases of adverse test results, by a full pharmacopoeial analysis. The book opens with a brief description of the importance of basic tests as one of the many steps needed to ensure a supply of safe and effective drugs. Chapter two describes several collections of more sophisticated tests, including volumetric or spectrophotometric analysis and thin-layer chromatography, that can be useful in the primary screening of imported pharmaceutical substances, and dosage forms. Information on how to obtain and use these guides to tests, which have not been published by WHO is also provided. Against this background, the main part of the book sets out test procedures for verifying the identity of selected pharmaceutical substances, pharmaceutical dosage forms, and medicinal plant materials. The book concludes with a cumulative index of test procedures described here and in the related WHO publications "Basic Tests for Pharmaceutical Substances" and "Basic Tests for Pharmaceutical Dosage Forms".
